<dcterms:abstract xml:lang="fr">Plusieurs complexes de CuII (CuIIL) peuvent subir un cycle rédox entre CuIIL et CuIL en présence d’agents réducteurs, tels que les thiols, dans l’environnement intracellulaire. L’O₂ agit comme un accepteur d’électrons, conduisant à la production d’espèces réactives de l’oxygène (ROS). L’efficacité catalytique dépend de la capacité du ligand à stabiliser à la fois CuII et CuI, évitant ainsi la libération de CuI en présence de chélateurs forts, tels que les métallothionéines. Cette thèse explore le rôle des thiols dans l’activité catalytique des complexes CuIIL. Trois classes de ligands ont été étudiées : le Dp44mT (une thiosemicarbazone α-N-hétérocyclique), une thiosemicarbazone dimérique, et une série de polypyridines tripodes (L0-3). Les résultats montrent que la stabilité seule ne garantit pas une bonne efficacité catalytique. Le CuIIDp44mT est très stable mais faiblement actif, probablement en raison de son faible potentiel rédox. En revanche, les CuIIL0–3, bien que moins stables, favorisent une oxydation plus importante des thiols. Les complexes les plus efficaces sont donc ceux qui assurent un bon équilibre entre réduction, réoxydation et rétention du CuI.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Several CuII complexes (CuIIL) can undergo a redox cycle between CuIIL and CuIL in the presence of reducing agents, such as thiols, in the intracellular environment. O2 acts as the electron acceptor, leading to reactive oxygen species (ROS) production. Catalytic efficiency depends on the ligand’s ability to stabilize both CuII and CuI, avoiding CuI release in the presence of strong chelators, such as metallothioneins. This thesis investigates the role of thiols in CuIIL catalytic activity. Three ligand classes were studied: Dp44mT (an α-N-heterocyclic thiosemicarbazone), a dimeric thiosemicarbazone, and a series of tripodal polypyridines (L0-3). Results show that stability alone does not ensure efficiency. CuIIDp44mT is highly stable but poorly active, likely due to its low redox potential. In contrast, CuIIL0-3 are less stable but promote greater thiol oxidation. Thus, the most effective complexes balance reduction, reoxidation, and CuI retention.</dcterms:abstract>
