<dcterms:abstract xml:lang="fr">Les hétérocycles mixtes azotés et soufrés constituent une famille de composés largement représentée parmi les médicaments, ce qui justifie l’intérêt porté au développement de nouvelles méthodes de synthèse rapides et efficaces pour accéder à de tels structures. Cette thèse a eu pour objectif la mise au point d’une plateforme soufrée-azotée, permettant l’accès rapide à une grande diversité d’hétérocycles (N,S). La stratégie mise en œuvre repose sur une séquence domino combinant deux réarrangements sigmatropiques conduisant à des 2-aminobuta-1,3-diènes. La réactivité de ces synthons a été explorée selon trois approches afin d’obtenir des hétérocycles (N,S):(I) une méthode divergente de sulfénylation intramoléculaire d’énamines menant à deux familles de benzo[4,5]imidazo[2,1-b]thiazoles chiraux; (II) une séquence domino d’additions nucléophiles pour l’obtention de dihydrothiazoles dans des conditions respectueuses de l’environnement; (III) l’hydroamination intramoléculaire de sulfures homallényliques et la cyclohydrocarbonylation de sulfures homoallyliques pour l’accès à différentes tailles de d’hétérocycles (N,S).</dcterms:abstract>
<dcterms:abstract xml:lang="en">Mixed nitrogen-sulfur heterocycles are among a major class of structural motifs in pharmaceuticals, which justifies the development of new, rapid and efficient synthetic strategies for accessing such structures.The present thesis focused on the design of a new N,S-platform, enabling fast access to a broad range of N,S-heterocycles. The approach relies on a domino sequence combining two successive sigmatropic rearrangements to generate sulfur-containing 2-aminobuta-1,3-dienes. The reactivity of these synthons was subsequently investigated through three main pathways, enabling the preparation N,S-heterocycles :(I) A divergent intramolecular sulfenylation of enamines, leading to two chiral benzo[4,5]imidazo[2,1 b]thiazoles families; (II) A domino nucleophilic addition process, providing access to dihydrothiazole derivatives under environmentally friendly conditions; (III) the intramolecular hydroamination of homoallenyl sulfides and the cyclohydrocarbonylation of homoallyl sulfides, to access different sizes of N,S-heterocycles. These results open perspectives for the synthesis of N,S-heterocycles with therapeutic potential.</dcterms:abstract>
