<dcterms:abstract xml:lang="fr">Les métastases, responsables de la majorité des décès par cancer, demeurent difficiles à traiter, en raison d’une pathogénèse très complexe. CDK5, joue un rôle clé dans la migration et l’invasion tumorale. Son inhibition perturbe la polarité cellulaire et réduit la dissémination métastatique. Malgré cela, aucun inhibiteur sélectif de CDK5 n’a atteint le stade clinique. Ce projet vise le développement de PROTACs ciblant CDK5 selon deux approches différentes : orthostérique, utilisant des inhibiteurs ATP-compétitifs hautement sélectifs, et allostérique, exploitant des modulateurs identifiés au laboratoire. Cette double stratégie permettra de caractériser le rôle de CDK5 dans la migration cellulaire et de valider le mécanisme allostérique de nos modulateurs via la dégradation de CDK5. Parallèlement, ce projet intègre également un axe dédié à la diversité moléculaire, avec la conception de nouveaux châssis, la 2,3-benzoxazépin-4-one et 1,2,5-triazépin-3-one.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Metastases, responsible for the majority of cancer-related deaths, remain difficult to treat due to a highly complex pathogenesis. CDK5 plays a key role in tumor cell migration and invasion. Its inhibition disrupts cell polarity and reduces metastatic dissemination. However, no selective CDK5 inhibitor has reached the clinical stage. This project aims to develop PROTACs targeting CDK5 via two different approaches: orthosteric, using highly selective ATP-competitive inhibitors, and allosteric, exploiting modulators identified in the laboratory. This dual strategy will allow the characterization of CDK5’s role in cell migration and the validation of the allosteric mechanism of our modulators via CDK5 degradation. Additionally, the project includes a molecular diversity axis, focusing on the design of new scaffolds, including 2,3-benzoxazepin-4-one and 1,2,5-triazepin-3-one.</dcterms:abstract>
