<dcterms:abstract xml:lang="fr">Le trouble dépressif majeur est associé à des perturbations des rythmes circadiens et de la plasticité synaptique dans des régions clés de la régulation de l’humeur, comme le cortex préfrontal médian (mPFC) et l’hippocampe (HC). À l’aide de modèles murins de dépression induite par le stress et d’altération ciblée de l’horloge circadienne dans les neurones excitateurs du mPFC, nous avons combiné des approches comportementales, moléculaires et électrophysiologiques. Le stress chronique perturbe l’horloge cérébrale et le sommeil, tandis que la kétamine et la privation de sommeil améliorent l’humeur via des mécanismes circadiens distincts. L’administration de modulateurs pharmacologiques de l’horloge induisent des effets variables selon le gène circadien ciblée. Ces effets passent par la régulation d’Homer1a, une protéine de plasticité synaptique influençant les récepteurs AMPA et la transmission glutamatergique, et conditionnant ainsi les effets anti- ou pro-dépresseurs observés.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Major depressive disorder is associated with disruptions in circadian rhythms and synaptic plasticity in key brain regions involved in mood regulation, such as the medial prefrontal cortex (mPFC) and hippocampus (HC). Using mouse models of stress-induced depression and targeted circadian clock disruption in excitatory neurons of the mPFC, we combined behavioral, molecular, and electrophysiological approaches. Chronic stress alters brain clock function and sleep architecture, while ketamine and sleep deprivation improve mood through distinct circadian mechanisms. The use of pharmacological clock modulators produces variable effects depending on the specific circadian component targeted. These effects are mediated, in part, through the regulation of Homer1a, a synaptic plasticity-related protein that modulates AMPA receptor expression and glutamatergic transmission, ultimately contributing to the observed pro- or antidepressant outcomes.</dcterms:abstract>
