<dcterms:abstract xml:lang="fr">L’objectif de ce doctorat était de développer des outils génétiques pour l’étude et la réduction de l’infection par les arbovirus chez le moustique Aedes aegypti. Nous avons conçu une stratégie de Forçage Génétique (FG) afin de renforcer l’immunité antivirale, fondée sur un système Toxine-Antidote (TA). La Toxine CRISPR-Cas9 cible le gène essentiel et haploinsuffisant lipophorine (lp), et l’antidote est une version recodée et résistante de lp. Chez les descendants de croisements entre FG et sauvages (WT), la mutation de l’allèle WT est létale, empêchant le croisement et conduisant à la suppression de la population WT au-delà d’un seuil de libération de moustiques FG. Un système TA fonctionnel a été établi chez A. aegypti et sera optimisé. En parallèle, une lignée transgénique a été développée pour l’étude de la salivation, révélant une absence de corrélation entre salivation et transmission virale, et une réduction de la salivation lors d’infections par les virus Zika ou chikungunya.</dcterms:abstract>
<dcterms:abstract xml:lang="en">This PhD aimed to develop genetic tools to investigate and mitigate arbovirus infection in the mosquito Aedes aegypti. First, a Gene Drive strategy was designed to enhance mosquito antiviral immunity through a CRISPR-Cas9 Toxin-Antidote (TA) system. The Toxin targets the essential, haploinsufficient lipophorin (lp) gene, while the Antidote encodes a recoded, resistant version of lp. In the progeny of Gene Drive (GD) and wild-type (WT) crosses, mutation of the WT lp allele is lethal, preventing interbreeding and enabling WT population suppression above a certain release threshold of GD mosquitoes. An efficient TA system was established in A. aegypti and is to be further optimized. In parallel, a transgenic reporter line was developed to study salivation. Results showed that salivation does not correlate with viral transmission and is significantly reduced upon Zika or chikungunya infection, challenging the current model linking transmission to infected saliva release.</dcterms:abstract>
