<dcterms:abstract xml:lang="fr">Cette thèse étudie comment la communication cérébello-préfrontale soutient la prédiction temporelle à travers plusieurs paradigmes comportementaux. Chez la souris maintenue, le réseau cérébello-préfrontal réorganise son activité selon le contexte de prédictibilité temporelle, la stimulation optogénétique du cervelet déclenchant des effets comportementaux et neuronaux dans le cortex prélimbique distincts entre les deux contextes. En condition libre, nous avons étendu l’étude aux échelles sub- et supra-secondes, montrant que les souris utilisent leurs attentes temporelles aux deux échelles et les mettent à jour de manière dynamique lorsque le contexte change. Enfin, les souris Synapsine II KO, modèle de schizophrénie, sont sélectivement altérées dans l’anticipation temporelle pour les aspects cognitifs du comportement. Bien que les marqueurs neuronaux de l’anticipation dans et entre le cervelet et le cortex préfrontal soient préservés, ils se dissocient du comportement. Ces résultats montrent que les boucles cérébello-préfrontales encodent de façon adaptative la structure temporelle et que leur altération pourrait sous-tendre certains déficits observés dans la schizophrénie.</dcterms:abstract>
<dcterms:abstract xml:lang="en">This thesis investigates how cerebello–prefrontal communication supports temporal prediction across multiple behavioral paradigms. In head-restrained mice, the cerebello–prefrontal network reorganized its activity according to the temporal predictability context, as optogenetic stimulation of the cerebellum triggered distinct behavioral and neuronal effects within the prelimbic cortex. In freely moving animals, we extended our investigation to both sub- and suprasecond ranges, showing that mice use temporal expectations across timescales and dynamically update them when the context changes. Finally, Synapsin II knockout mice, a model for schizophrenia, were selectively impaired in using temporal expectations for higher-order aspects of goal-directed behavior. Although neural markers of anticipation within and between the cerebellum and prefrontal cortex remained present, they decorrelated from behavioral outcome. Together, these results show that cerebello–prefrontal loops adaptively encode temporal structure, and their alteration may underlie timing and predictive deficits in schizophrenia.</dcterms:abstract>
