<dcterms:abstract xml:lang="fr">Le vieillissement est un facteur de risque cardiovasculaire majeur conduisant à l’insuffisance cardiaque ; cette étude a évalué si la combinaison de procyanidines (GSE) et d’oenothéine B (OTB) apporte des bénéfices cardiovasculaires. Des rats jeunes (12 semaines) et d’âge moyen (15 mois), non traités ou traités 2 semaines par GSE (30 mg/kg/j), OTB (10 mg/kg/j) ou GSE+OTB, ont été évalués pour la pression artérielle, les fonctions cardiaque et vasculaire, le stress oxydant et des marqueurs moléculaires. Les rats d’âge moyen présentaient hypertension systolique, dysfonction diastolique, hypertrophie ventriculaire gauche, fibrose, inflammation, stress oxydant et vasoconstriction microvasculaire. L’augmentation des ROS impliquait NOS et la voie AT1R/NADPH oxydases/SGLT1/2. Ces altérations étaient inversées par GSE+OTB et partiellement par chaque composé seul, suggérant que des combinaisons définies de polyphénols sont prometteuses pour un vieillissement cardiovasculaire sain.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Ageing is a major cardiovascular risk factor leading to heart failure; this study evaluated whether combining procyanidins (grape seed extract, GSE) and oenothein B (OTB) provides cardiovascular benefits. Young (12 weeks) and middle-aged rats (15 months), untreated or treated for 2 weeks with GSE (30 mg/kg/day), OTB (10 mg/kg/day) or GSE+OTB, were assessed for blood pressure, cardiac and vascular function, oxidative stress and molecular markers. Middle-aged rats developed systolic hypertension, diastolic dysfunction, left ventricular hypertrophy, fibrosis, inflammation, oxidative stress and microvascular vasoconstriction. Increased ROS formation in the left ventricle and resistance arteries involved NOS and the AT1R/NADPH oxidases/SGLT1/2 pathway. The left ventricle and aortic arch showed markers of inflammation, endothelial activation, remodeling and fibrosis. These alterations were reversed by GSE+OTB and partially by each compound alone, supporting defined polyphenol combinations as a strategy for healthy ageing.</dcterms:abstract>
