<dcterms:abstract xml:lang="fr">L’exercice physique, en tant que pilier dans la prise en charge des patients atteints de DT2, représente une source prometteuse de traitements principalement via la sécrétion d’exerkines. L’objectif de cette étude a consisté, dans un premier temps, à étudier l’impact d’une exerkine d’intérêt, ILV-001, sur un modèle de pré-adipocytes humains différenciés, nécessitant la mise au point d’un modèle in vitro en 2D. Les premiers résultats obtenus suggèrent qu’ILV-001 pourrait agir de façon dose-dépendante. L’évaluation de l’impact de l’exerkine en fonction du contexte métabolique ne révèle pas d’impact en condition physiologique. Des prélèvements supplémentaires issus de donneurs DT2 permettraient de mieux définir son rôle en condition pathologique. Par ailleurs, l’analyse des sécrétions adipocytaires post-traitement par ILV-001 propose un effet différentiel en fonction du contexte métabolique. Nous avons ainsi cherché à déterminer si ces sécrétions pouvaient influencer la santé des îlots pancréatiques humains. En résumé, ce travail a permis de contribuer à la compréhension des mécanismes sous-jacents aux bienfaits de l’exercice physique via un intérêt porté sur les exerkines en particulier ILV-001.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Physical exercise, one of the first recommendation for patient care, represents a promising treatment attributed to the secretion of exerkines. The first objective of this study was to investigate the impact of ILV-001, an exerkine of interest, in adipocyte functions, involving the development of a 2D in vitro model of differentiated human pre-adipocytes derived from primary culture. The first results suggest that ILV-001 may act in a dose-dependent manner, with a positive impact on insulin sensitivity observed at low concentrations. Assessing the impact of the exerkine with respect to metabolic status does not reveal any effect under physiological conditions. Additional samples from T2D donors would help to better define its role in a pathological context. Analysis of adipocyte secretions following ILV-001 treatment suggests a differential effect depending on the metabolic context. Thus, we aimed to determine whether these secretions could influence the health of human pancreatic islets. This research has enabled an in-depth understanding of the mechanisms underlying the benefits of physical exercise through the action of the exerkine ILV-001.</dcterms:abstract>
