<dcterms:abstract xml:lang="fr">Cette thèse étudie l’expression et le rôle fonctionnel du récepteur du GLP-1 (GLP-1R) dans le système cardiovasculaire humain. Une revue exploratoire a cartographié la distribution vasculaire du GLP-1R selon les territoires, types cellulaires et états pathologiques. Une étude translationnelle sur tissus humains a analysé l’expression du GLP-1R et sa modulation par l’inflammation. Les résultats montrent que l’inflammation augmente l’expression et modifie la localisation du GLP-1R, principalement vers le compartiment musculaire lisse, et que son activation réduit le stress oxydant via la voie AMPc/PKA. Ces données apportent un éclairage mécanistique aux effets vasculaires protecteurs des agonistes du GLP-1R et soutiennent leur potentiel thérapeutique dans les maladies cardiovasculaires inflammatoires.</dcterms:abstract>
<dcterms:abstract xml:lang="en">This thesis investigates the expression and functional role of the GLP-1 receptor (GLP-1R) in the human cardiovascular system. A scoping review mapped vascular GLP-1R distribution across vascular beds, cell types, species, and pathological states. A translational study on human cardiovascular tissues assessed GLP-1R expression and its modulation by inflammation. Results show that inflammatory stimuli upregulate GLP-1R expression and alter its spatial localization, with redistribution toward the vascular smooth muscle layer. GLP-1R activation reduced oxidative stress via the cAMP/PKA signaling pathway. These findings provide mechanistic insight into the vascular protective effects of GLP-1 receptor agonists and support their therapeutic potential in inflammatory cardiovascular diseases.</dcterms:abstract>
