<dcterms:abstract xml:lang="fr">Un déséquilibre de la balance énergétique, associé à une alimentation riche en graisses, favorise l’obésité. La leptine, hormone anorexigène produite par le tissu adipeux, est abondante chez les personnes obèses, mais pourtant son message de satiété n’est plus perçu, traduisant une résistance à l’hormone. Les mécanismes et acteurs de cette résistance restent encore mal compris. Mes travaux de thèse montrent que les microglies pourraient jouer un rôle clé dans cette régulation. Après avoir été internalisée par les microglies, la leptine transite dans différents compartiments du trafic intracellulaire avant d’être relarguée. Elle induit également la sécrétion de cytokines anti-inflammatoires impliquées dans l’homéostasie neuronale. En revanche, l’exposition à l’acide palmitique ralentit ce relargage et favorise la production de cytokines pro-inflammatoires. Ces résultats révèlent une nouvelle fonction des microglies dans l’homéostasie énergétique et ouvrent la voie à de nouvelles approches pour inverser la résistance à la leptine.</dcterms:abstract>
<dcterms:abstract xml:lang="en">An imbalance in energy homeostasis, associated with a high-fat diet, promotes obesity. Leptin, an anorexigenic hormone produced by adipose tissue, is highly abundant in obese individuals, yet its satiety signal is no longer perceived, reflecting a resistance to the hormone. The mechanisms and cellular actors underlying this resistance remain poorly understood. My doctoral work shows that microglial cells may play a key role in this regulation. After being internalized by microglia, leptin travels through different compartments of the intracellular trafficking pathway before being released. It also induces the secretion of anti-inflammatory cytokines involved in neuronal homeostasis. Conversely, exposure to palmitic acid slows this release and promotes the production of pro-inflammatory cytokines. These findings reveal a new function for microglia in energy homeostasis and open the way to new approaches for reversing leptin resistance.</dcterms:abstract>
