<dcterms:abstract xml:lang="fr">Les protéines ATF7 sont des protéines à leucine-zipper caractérisées par leur capacité à se fixer sur les séquences ATF/CRE (TGACGTCA) de différents promoteurs précoces d'adénovirus et de certains gènes cellulaires. Elles sont capables d'interagir avec certains membres de la famille des oncoprotéines Jun/Fos et de moduler leurs activités. Pour préciser le rôle d'ATF7 dans les mécanismes d'activation transcriptionnelle, nous avons analysé ses relations avec les hsTAFs, des protéines qui font partie de plusieurs complexes multiprotéiques comme le complexe TFIID. Nos résultats indiquent que l'activité transcriptionnelle d'ATF7 est spécifiquement relayée par hsTAF12, principalement par l'isoforme de 20 kDa. De plus ATF7 et hsTAF12 interagissent in vivo et l'intégrité du domaine activateur amino-terminal d'ATF7 et du motif "histone-fold" de hsTAF12 sont indispensables à cette interaction. Nous montrons enfin que la transactivation relayée par hsTAF12 est spécifiquement inhibée par hsTAF4 (son partenaire d'hétérodimérisation au sein de TFIID) mais pas par hsTAF4b, un homologue tissu-spécifique de hsTAF4.Parmi les protéines interagissant et modulant l'activité d'ATF7, notre équipe a détecté la protéine Ubc9, une enzyme clé de la voie de SUMOylation. Par ailleurs, nous avons identifié dans la partie amino-terminale d'ATF7 un motif consensus d'un site de SUMOylation et montré que la protéine est SUMOylée, à la fois par des expériences in vitro et in vivo. Nous avons observé que la localisation intracellulaire d'ATF7 était dépendante de sa SUMOylation : nos résultats suggèrent qu'ATF7 ne serait présent dans le noyau qu'à l'état dé-SUMOylé ; la SUMOylation cytoplasmique d'ATF7 induirait sa séquestration au niveau des complexes des pores nucléaires (NPC). Nous avons notamment mis en évidence, par des expériences d'immunomarquage, une colocalisation d'ATF7 avec une protéine des NPC, RanBP2. Cette dernière est également une enzyme E3 de la voie de SUMOylation qui augmente spécifiquement la SUMOylation d'ATF7 in vitro. De plus, une protéine ATF7 non SUMOylable (mutée au niveau de son site unique de SUMOylation) présente une activité transcriptionnelle nettement supérieure à celle de la protéine SUMOylée. Des expériences d'immunoprécipitation de chromatine (ChIP) ont également révélé que la SUMOylation d'ATF7 diminue sa présence au niveau de ses promoteurs cibles.</dcterms:abstract>
<dcterms:abstract xml:lang="en">ATF7 proteins, which are members of the b-Zip family of transcription factors, are able to bind ATF/CRE sequences (TGACGTCA) within different early adenoviral or cellular promoters. ATF7 can interact with the family of Jun/Fos oncoproteins and modulate their activities. To get an insight into the transactivation mechanism mediated by ATF7, we analyzed its relations with hsTAFs proteins, which are components of several multiproteic complexes like TFIID. Our results show that transactivation by ATF7 is specifically mediated by hsTAF12, mainly by its 20-kDa isoform. Moreover, ATF7 and hsTAF12 interact in vivo : the integrity of the ATF7 amino-terminal activation domain and of the hsTAF12 "histone-fold" domain is required for this interaction. We show that hsTAF12 mediated transactivation is specifically inhibited by hsTAF4 (its heterodimerization partner within TFIID), and not by hsTAF4b, a tissue-specific hsTAF4 homolog.Ubc9, a protein involved in the SUMOylation pathway, was found among the proteins interacting with ATF7 and modulating its activity. We have identified a consensus SUMOylation site within the N-terminal part of ATF7 and demonstrate that ATF7 is SUMOylated both by in vitro and in vivo experiments. Moreover, our results reveal that the intracellular localization of ATF7 is affected by its SUMOylation : ATF7 is found in the nucleus only in a de-SUMOylated form ; the cytoplasmic SUMOylation of ATF7 likely induces its sequestration at the level of the nuclear pore complexes (NPC). Using immunohistochemistry assays, we observe a colocalization between ATF7 and RanBP2, a protein of the NPC, which is an E3 ligase of the SUMOylation pathway. Accordingly, RanBP2 is able to stimulate ATF7 SUMOylation in vitro. This NPC targeting seems to influence ATF7 transactivation activity as an ATF7 mutant, whose SUMOylation is impaired, is more active than its SUMOylated counterpart. These results correlate with our chromatin-immunoprecipitation (ChIP) experiments, which show that the occupancy of a target promoter by ATF7 is highest in the presence of the unSUMOylated protein.</dcterms:abstract>
