<dcterms:abstract xml:lang="fr">L'exocytose est un processus cellulaire primordial qui permet aux cellules d'assurer leurs fonctions spécifiques en sécrétant une multitude de signaux chimiques extrinsèques tels que les hormones, les neurotransmetteurs, les cytokines ou encore les facteurs de croissance. Dans les cellules neuroendocrines, l'exocytose requiert une réorganisation fine et précise du cytosquelette d'actine, phénomène étroitement régulé par certaines GTPases monomériques de type Rho, dont Cdc42 localisée à la membrane plasmique. Cependant le mécanisme par lequel Cdc42 régule la dynamique d'actine ainsi que le rôle exact de l'actine au cours de l'exocytose restaient largement incompris. Le but majeur de mon projet de thèse a été d'élucider d'une part, les voies effectrices par lesquelles Cdc42 contrôle la sécrétion d'hormones dans les cellules neuroendocrines et d'autre part, d'identifier les protéines nécessaires à la régulation de son activité (protéines GEF et GAP) au cours de l'exocytose. En étudiant la sécrétion neuroendocrine dans les cellules chromaffines et PC12, j ai pu montrer lors de mon doctorat que Cdc42 contrôle l'exocytose en activant une cascade moléculaire impliquant la protéine N-WASP et le complexe Arp2/3 localisé sur les granules de sécrétion. Cette cascade aboutit à une synthèse de novo d'actine indispensable aux étapes tardives de la sécrétion. J ai ensuite montré que l'activité de Cdc42 lors de l'exocytose est contrôlée par l'intersectine-1L, un facteur d'échange spécifique de cette GTPase impliqué jusqu alors principalement dans les phénomènes d'endocytose. L'ensemble de ces données nous a amené à suggérer un processus inédit au sein duquel le cytosquelette d'actine n est plus cantonné à un simple rôle de barrière physique mais assure également un rôle actif dans les étapes tardives de l'exocytose. De plus, la découverte de l'intersectine1-L'en tant que facteur d'échange activant Cdc42 n est pas moins novatrice puisqu elle constitue la première démonstration d'une implication fonctionnelle de l'intersectine au cours d'un processus d'exocytose.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Through releasing numerous chemical molecules as hormones, neurotransmitters, cytokines or growth hormone, exocytosis is one of the major processes allowing cells to provide their specific functions. Most secretory cells undergoing calcium-regulated exocytosis in response to cell surface receptor stimulation display a dense subplasmalemmal actin network, which is remodeled during the exocytotic process. Rho GTPases are well-known regulators of actin dynamics, and membrane-located Cdc42 was thought to play a role in regulated exocytosis. However, the exact role of actin during exocytosis and the pathway used by Cdc42 to regulate actin dynamics were misunderstood. The main project of my thesis was to decipher the effector pathways by which Cdc42 controls hormone release and to identify the proteins responsible for its regulation (GAP and GEF) during exocytic activity. By studying neuroendocrine secretion in chromaffin and PC12 cells, I have demonstrated that secretagogue-evoked stimulation induces the sequential ordering of Cdc42, N-WASP and Arp2/3 at the interface between granules and the plasma membrane, thereby providing a way to specifically target local actin filament polymerization at granule docking sites. This is the first time a molecular support for the de novo formation of actin filaments during the course of exocytosis has been described. Then I demonstrated that intersectin-1L exchange factor, first implicated in endocytosis, is responsible for Cdc42 activation during the exocytotic process. Taken together, these datas suggest an original process where actin is not only a physical barrier that hinders the movement of granules but presents an active role during the late steps of exocytosis. Moreover, discover of intersectin-1L as Cdc42 exchange factor is the first demonstration for the functionnal implication of this protein in endocytic process.</dcterms:abstract>
