<dcterms:abstract xml:lang="fr">Les procyanidols (Pcy) indue l'apoptose des cellules cancéreuses coliques mais les mécanismes sont inconnues. Nous étudions les mécanismes d'action des Pcy de la pomme dans les cellules cancéreuses coliques humaines SW480 et SW620. Le MDL'renforce les effets proapoptotiques des Pcy dans les cellules SW620 mais non dans les cellules SW480. Pcy/MDL'provoquent une chute du contenu intracellulaire des polyamines et l'activation de la voie extrinsèque TRAIL-DR4/DR5 dans les cellules SW620. Les Pcy activent la voie mitochondriale dans les cellules SW620 associé à la production de radicaux libres. Les Pcy augmentent la sensibilité à TRAIL'dans cellules SW480, et inhibent la résistance au TRAIL'pour les SW620. Les Pcy activent DR4/-DR5 indépendant des lipid-rafts. Les Pcy sensibilisent les SW480 à la voie Fas. Par contre les SW620 sont résistent à Fas, malgré l'augmentation de l'expression de Fas en présence de Pcy. L'activation du Fas dans les SW620 se produit après un blocage des DR4/DR5.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Apple procyanidins (Pcy) trigger apoptosis in colon cancer cells but the mechanisms are unknown. We studied apoptotic mechanisms triggered by apple Pcy in human colon cancer SW480 and SW620 cells. We showed that MDL, inhibitor of PAO enzyme, in SW620 cells switched mitochondrial pathway activated by Pcy to TRAIL-DR4/DR5-mediated apoptosis, related to depletion of intracellular polyamine. In SW480 cells the proapoptotic effects of Pcy were similar to those with Pcy/MDL. Pcy enhanced sensitivity of SW480 cells to TRAIL and overcome TRAIL-resistance in SW620 cells by activating caspase-dependent apoptosis. Pcy-triggered apoptosis in SW620 cells involved a cross-talk between extrinsic and intrinsic pathways, with ROS production favoring mitochondria disruption. DR4/DR5 activation was independent of lipid-raft. We showed that Pcy activated Fas in SW480 cells, but not in SW620 cells despite up-regulated Fas expression. Activation of Fas by Pcy occurred in SW620 cells after block DR4/DR5.</dcterms:abstract>
