<dcterms:abstract xml:lang="fr">Les prokinéticines sont une famille de protéines sécrétées et classées en deux types, la prokinéticine-1 et la prokinéticine-2. Les prokinéticines sont des nouveaux facteurs angiogéniques qui exercent leurs fonctions biologiques via deux récepteurs couplés aux protéines G : PKR1 et PKR2. Ils sont exprimés dans divers organes et notamment dans le coeur des souris et de l'homme. L'objectif de mon projet de thèse était de déterminer la contribution de la signalisation de PK-2/PKR1 dans les cellules endothéliales cardiaques et les cellules dérivées de l'épicarde lors de la néovascularisation. Les travaux de recherche sur le rôle du récepteur PKR1 dans les cellules endothéliales dérivées du coeur ont permis d'identifier une nouvelle voie de signalisation lors du processus de l'angiogenèse cardiaque. La prokinéticine-2 via PKR1 stimule la prolifération et la migration des cellules endothéliales H5V dérivées du coeur des souris sans l'intervention de PKR2. En outre, le récepteur PKR1 couplé à la protéine Ga11 est responsable de l'activation de l'angiogenèse in vitro non pas en régulant l'expression du facteur VEGF mais en activant directement les protéines kinases MAPK et PI3K. D autre part, pour la première fois ces études montrent qu un récepteur couplé aux protéines G, PKR1 active la prolifération et la différenciation des cellules dérivées de l'épicarde (EPDC) en cellules vasculaires (cellules endothéliales et cellules musculaires lisses) aux dépens des fibroblastes. Ainsi au stade post-natal et au stade adulte la signalisation de PKR1 permet de restaurer la multipotence des cellules EPDC et d'orienter leur différenciation en cellules vasculaires. L'angiogenèse et la reprogrammation des cellules souches EPDC en cellules vasculaires ont un intérêt majeur notamment lors de la néovascularisation. Par conséquent, PKR1 semble être une cible potentielle pour régénérer la vascularisation et traiter ainsi les cardiopathies ischémiques.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Prokineticins are a family of secreted peptides, comprising two classes: prokineticin-1 (PK-1) and prokineticin-2 (PK-2). Prokineticins are new angiogenic factors and exert their biological activity by binding to two G protein-coupled receptors: PKR1 and PKR2. They are quite widely expressed in mammalian tissues and organs, notably in mouse and human heart. The purpose of my thesis was to investigate the contribution of cardiac endothelial cells and epicardial-derived progenitor cells into prokineticin-2/PKR1-mediated neovascularization. The research work on PKR1 function in endothelial cell (H5V) derived from heart lead us to identify a new signalling pathway during angiogenesis. Activation of the PKR1 signaling pathway via PK-2 stimulates proliferation, migration and angiogenesis in endothelial cells (H5V). Moreover in cultured cardiac endothelial cells, stimulation of PKR1 coupled to Ga11 protein induces vessel-like formation without increasing VEGF levels. PKR1 signaling directly induces angiogenesis, activating MAPK and PI3K. On the other hand, for the first time in vitro studies reveal that activation of PKR1 promotes epicardial-derived progenitor cells (EPDC) proliferation and differentiation into endothelial and smooth muscle cells. Besides, prokineticin-2/PKR1 signaling can reprogram adult EPDC to adopt a vasculogenic cell fate at the expense of fibroblast. I found that prokineticin-2/PKR1 signaling cell-autonomously activates endothelial cells proliferation, migration and angiogenesis. EPDC differentiation into vasculogenic cells by prokineticin-2/PKR1 signaling is a new pathway to stimulate post-natal neovascularization. Since angiogenesis and stem cell differentiation into vasculogenic cells are two significant process during neovascularization, PKR1 could be a new potential therapeutic target to regenerate vascularization for the treatment of ischemic heart diseases.</dcterms:abstract>
