<dcterms:abstract xml:lang="fr">Les statines sont des médicaments très efficaces dans le traitement de l'hypercholestérolémie. Cependant, elles entraînent des effets délétères qui se traduisent par l'apparition de douleurs musculaires. Il a été clairement établi que la mitochondrie était une cible privilégiée des statines et était la principale source des radicaux libres. Cependant, aucune étude n a été consacrée sur l'effet de statines sur cette production de radicaux libres. La première partie de ma thèse a démontré que, in vitro, l'atorvastatine diminue la respiration mitochondriale musculaire, quel que soit le phénotype, et augmente la production de ROS. Par contre, un entraînement en endurance prévient ces effets délétères sur les muscles squelettiques. Par ailleurs, nous avons démontré que, sur des rats traités chroniquement avec l'atorvastatine, les capacités d'endurance sont diminuées. La seconde partie de ma thèse a montré que ces statines agissent selon le concept du mitohormesis . En effet, elles entraînent dans le muscle squelettique une augmentation importante de radicaux libres débordant le système antioxydant et entraînant une désactivation de la biogenèse mitochondriale et une altération des capacités oxydatives musculaires. Par contre, dans le cœur où les capacités antioxydantes sont importantes, elles entraînent une augmentation modérée des ROS qui permet l'activation de voies de signalisation menant à la biogenèse mitochondriale. Ces travaux ont permis de comprendre pourquoi et comment les statines agissaient différemment dans le cœur et les muscle squelettiques et d'ouvrir des pistes dans le traitement des douleurs musculaires suite à un traitement avec des statines.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Statins is very effective drugs in the treatment of hypercholesterolemia. However, statins induced deleterious effects including muscular pains. It was clearly established that mitochondria were a privileged target of statins and were the principal source of fre radicals. However, no study concerned the effect of statins on this ROS production. The first part of my thesis demonstrated that in vitro, atorvastatin decreased mitochondrial respiration whatever the phenotype and increased the ROS production. On the other hand, a chronically endurance training decreased these deleterious effects in skeletal muscles. Besides, we demonstrated that on treated rats chronically with atorvastatin, the endurance capacities decreased. The second part of my thesis showed that statins acted according to the concept of mitohormesis . Indeed, statins induced in glycolytic muscle an important increase of ROS extending beyond the antioxidant system inducing a deactivation of the mitochondrial biogenesis and a decrease of muscular oxidative capacities. On the other hand, in the cardiac muscle where the antioxidant capacities are important, statins induced a moderate increase of ROS which allowed to the activation of the mitochondrial biogenesis. These works allowed understanding why and how statins acted differently in cardiac and skeletal muscles and opening perspectives in the treatment of the muscular pains following a treatment with statins.</dcterms:abstract>
