<dc:description xml:langue="" xsi:type="unistra:Resume">Le lupus érythémateux systémique ￼(LES) est une maladie auto-immune, chronique, complexe et systémique, marquée par une grande hétérogénéité, clinique et biologique. Cette thèse développe les mécanismes immunopathologiques du ￼LES, notamment l’implication des cellules B, des interférons de type 1 et des défauts de clairance apoptotique dans la rupture de la tolérance immunitaire. ￼Une large place est accordée à l’analyse comparative des approches thérapeutiques : traitements conventionnels, biothérapies actuellement disponibles (belimumab, anifrolumab), et stratégies émergentes en cours d’étude. L’évaluation critique de ses options permet de mieux cerner leurs indications, limites et perspectives. L’étude met également en lumière l’impact du lupus sur la qualité de vie, insistant sur l’importance d’une prise en charge globale et personnalisée.</dc:description>
<dc:description xml:langue="" xsi:type="unistra:Resume">Systemic lupus erythematosus (SLE) is a chronic, complex, and systemic autoimmune disease, characterized by significant clinical and biological heterogeneity. This thesis provides an analysis of the immunopathological mechanisms of SLE, focusing on the role of B cells, type 1 interferons, and defective apoptotic clearance in the breakdown of immune tolerance. Particular emphases is placed on a comparative review of therapeutic approaches: conventional treatments, currently available biologics (belimumab, anifrolumab), and emerging strategies under investigation. A critical assessment of these options highlights their respective indications, limitations, and future prospects. The study also addresses the impact of lupus on patients’ quality of life, underscoring the importance of a comprehensive and personalized care approach.</dc:description>
