Résumé(s) : Les travaux du laboratoire ont montré que la protéine Wnt5a protège contre l’athérosclérose en inhibant l’accumulation intracellulaire de cholestérol. Cependant, son usage thérapeutique direct est peu envisageable. Un criblage phénotypique de 7000 composés a permis d’identifier trois petites molécules inédites capables d’inhiber cette accumulation in vitro. L’une d’elles, nommée « Trafic 8 », s’est révélée particulièrement prometteuse par son action robuste et concentration-dépendante, sans toxicité apparente. Sa cible moléculaire et certaines voies de signalisation ont été identifiées, éclairant son mécanisme d’action. Trafic 8 présente aussi un bon potentiel d’optimisation chimique, pouvant améliorer son efficacité. Cette stratégie innovante, indépendante de la voie du récepteur LDL, pourrait compléter les traitements actuels et représenter une avancée majeure contre l’athérosclérose., The Wnt5a protein protects against atherosclerosis by reducing intracellular cholesterol and lesion formation. Its direct therapeutic use is limited, but pharmacological compounds mimicking its effects offer a promising alternative. A high-throughput phenotypic screen of 7,000 compounds identified three novel molecules that reduce cholesterol accumulation in vitro. This thesis focused on these candidates. The most promising, “Trafic 8,” showed strong, concentration-dependent inhibition of cholesterol buildup without in vitro toxicity. Its molecular target and associated signaling pathways were identified, clarifying its mechanism of action. Trafic 8 also displays high potential for chemical optimization to improve efficacy at lower doses. This innovative approach, independent of the LDL receptor pathway, could complement current therapies and represent a major advance in atherosclerosis prevention and treatment.
