Role of PGRPs during the activation and the control of the Drosophila melanogaster immune response

Cette thèse porte sur le rôle des PeptidoGlycan Recognition Proteins au cours de la réponse immunitaire innée de la drosophile. Mes résultats démontrent que PGRP-SD coopère avec PGRP-SA et GNBP-1 dans la reconnaissance des bactéries à Gram-positif et l’activation de la voie immune Toll, contrôlant l’induction de certains peptides antimicrobiens. J’ai ensuite démontré que PGRP-SC1 et SC2, qui ont une activité enzymatique amidase, régulent activement la voie IMD. En dégradant le peptidoglycane, ces amidases diminuent en effet son pouvoir immuno-stimulateur. L’absence de contrôle de la réponse immunitaire à des effets développementaux, probablement liés à l’augmentation de l’apoptose. Un travail préliminaire sur PGRP-LF, indique que cette protéine fonctionne comme régulateur négatif des voies IMD et JNK. PGRP-LF agit en l’absence et en présence d’une infection, en limitant l’activation de ces deux voies de signalisation au cours du développement.
I have studied the role of PeptidoGlycan Recognition Proteins during drosophila innate immunity. I have demonstrated that PGRP-SD cooperates with PGRP-SA and GNBP-1, in the recognition of various Gram-positive bacteria and the downstream signaling trough the Toll immune pathway, inducing antimicrobial peptide gene transcription. I next have shown that PGRP-SC1 and SC2, which have amidase activity, play a crucial role in down-regulating the IMD pathway. We propose that, by cleaving peptidoglycan, these amidases decrease its stimulatory potential. In the absence of such a regulation, an uncontroled immune response can lead to death or to developmental defects probably due to massive cell death. Preliminary results on the role of PGRP-LF indicate that this protein is a negative regulator both for the IMD immune pathway and for the JNK pathway. PGRP-LF can act in an infection independent and/or dependent manner, to limit the activation of these two pathways during the development.
